Production of Extracellular Water-Soluble Polysaccharides and Hydrolytic Enzymes from Pedobacter sp. PR-M6 for Fermentation

Abstract
Winter mushroom (Flammulina velutipes) substrate was used to investigate the characteristics of extracellular water-soluble polysaccharides (WSPs) and hydrolytic enzymes produced by Pedobacter sp. PR-M6. The dry weight of the relative insoluble substrate (IS) was found to increase continuously when cultured at 30¡É compared with that at 20¡É until day 7 of incubation. Three chitinase isozymes and one chitin deacetylase were identified by SDS-PAGE after 7 days of PR-M6 incubation. Analysis of the PR-M6 culture medium containing 1% winter mushroom powder indicated higher levels of ¥â-1,3-glucanase activity when cultured at 20¡É than when cultured at 30¡É. Dry weight of WSPs was the highest at 380 mg/100 mL in the mushroom medium at 20¡É incubation. The WSP production efficiency was the highest at 1% mushroom substrate. Yields of WSPs were 67.83 mg/100 mL and 111.57 mg/100 mL in fresh matter and dry matter, respectively. WSP yield showed no significant difference between pH 3 and pH 8.
